摘要:
:为探明草原革蜱和森林革蜱不同饱血状态下中肠菌群结构的特点,本研究在无菌条件下分别采集
饱血、半饱血草原革蜱和森林革蜱中肠内容物,提取细菌总DNA,PCR扩增细菌16S rRNA V3区,变性梯度凝胶
电泳(DGGE)后,切取优势条带纯化、克隆和测序,测序结果与GenBank数据库中已知细菌序列进行比对分析。
结果显示,在饱血、半饱血草原革蜱和森林革蜱中肠4个肠内容物样品中共检测到巴氏立克次体暂定种、柯克斯
体科菌、大肠杆菌、嗜吞噬细胞无形体和斑疹伤寒立克次氏体5种细菌,其中半饱血草原革蜱中肠内存在的菌种
为:巴氏立克次体暂定种、柯克斯体科菌和大肠杆菌;饱血草原革蜱中肠内存在的菌种为:巴氏立克次体暂定
种、柯克斯体科菌和嗜吞噬细胞无形体;半饱血森林革蜱中肠内存在的菌种为:巴氏立克次体暂定种、大肠杆
菌和嗜吞噬细胞无形体;饱血森林革蜱中肠内存在的菌种为:巴氏立克次体暂定种、柯克斯体科菌、嗜吞噬细
胞无形体和斑疹伤寒立克次氏体;4个样品中的共有菌为巴氏立克次体暂定种。研究结果表明,不同蜱种中肠内
菌群的结构随吸血时间的延长而发生变化。本研究将为防治草原革蜱、森林革蜱及相关蜱传病提供科学依据。
Abstract:
To investigate the characteristics of microflora in midgut of different engorged Dermacentor nuttalli and
Dermacentor silvarum, the midgut contents of different engorged Dermacentor nuttalli and Dermacentor silvarum were collected
under aseptic conditions. The total DNA of bacteria were extracted, and the V3 areas of 16S rRNA were amplified by PCR. After
denaturing gradient gel electrophoresis (DGGE), the predominant bands were cut for purification, cloning and sequencing. The
sequencing results were compared and analysed with the known bacterial sequences in GenBank. The results showed that 5
bacteria species including Candidatus Rickettsia barbariae, Coxiellaceae bacterium, Escherichia coli, Anaplasma phagocytophilum
and Rickettsia typhi were detected in four midgut samples of Dermacentor nuttalli and Dermacentor silvarum. Candidatus
Rickettsia barbariae, Coxiellaceae bacterium and Escherichia coli existed in half engorged midguts of Dermacentor nuttalli.
Candidatus Rickettsia barbariae, Coxiellaceae bacterium and Anaplasma phagocytophilum existed in fully engorged midguts of Dermacentor nuttalli. Candidatus Rickettsia barbariae, Escherichia coli and Anaplasma phagocytophilum existed in half engorged
midguts of Dermacentor silvarum. Candidatus Rickettsia barbariae, Coxiellaceae bacterium, Anaplasma phagocytophilum and
Rickettsia typhi existed in fully engorged midguts of Dermacentor silvarum. The common bacteria in the 4 samples were
Candidatus Rickettsia barbariae. The results showed that the microflora in midgut of different ticks changed with the prolonged
blood-feeding time. This study will provide the scientific basis for preventing and controlling the Dermacentor nuttalli and
Dermacentor silvarum, and related tick-borne diseases.
