摘要:
:为了解山东地区猪圆环病毒2型(PCV2)流行病学特点和遗传变异情况,本研究利用PCR方法对从
无害化处理场和屠宰场采集的1 477 份猪的组织样品进行了PCV2 检测和部分PCV2 全基因组克隆分析。结果显
示,1 477份样品中,有122份样品为PCV2阳性,阳性率为8.26%。选取22株进行病毒全基因组扩增,经过序列
比对和遗传进化分析显示,22株病毒分别为PCV2a(5株)、PCV2b(7株)、PCV2d(3株)和PCV2e(7株),表明山
东地区PCV2流行基因型具有多样性,并且以PCV2b和PCV2e流行为主。与其他基因型相比,PCV2优势基因型
PCV2b和PCV2e ORF2的氨基酸位点变化分别集中在59K、190T和47S、63T、130F、131P、133S、197I、191K位点,其中优势
基因型PCV2e与LG、DBN-SX07-2、SH和ZJ这4株疫苗株的Cap蛋白抗原指数差异最大,提示了疫苗可能对山
东省当前流行的PCV2免疫效果不理想。本研究结果可为后续PCV2分子生物学研究及疫苗的选用提供参考依据。
Abstract:
In order to investigate molecular epidemiology and genetic variation of porcine circovirus type 2 (PCV2), a total of
1 477 samples were collected from harmless disposal field and slaughter houses and then subjected to PCV2 detection. The results
showed that the positive rate of PCV2 was 8.26% (122/1 477). Twenty-two PCV2 positive samples were selected for complete
genome sequence analysis. Multiple sequence alignment in phylogenetic analysis results showed that 5 of 22 PCV2 positive
samples belonged to PCV2a subgenotype, 7 of 22 belonged to PCV2b subgenotype, 3 of 22 belonged to PCV2d subgenotype, and
7 of 22 belonged to PCV2e subgenotype. These data indicate that the genetic diversity of PCV2 was complex in Shandong
province, and the dominant subgenotype was PCV2b and PCV2e. Based on the analysis of ORF2 acid amino sequences, the
