:为筛选适用于新型猪源链球菌(SS)灭活疫苗的佐剂,本研究以SS1 Z1株、SS2 Z2株和SS7 Z7株为
抗原,分别配制ISA201 VG双相油乳佐剂疫苗(简称ISA201疫苗)、GEL 02 PR水溶性聚合物佐剂疫苗(简称GEL
疫苗)、IMS1313N VG水溶性纳米佐剂疫苗(简称IMS1313疫苗)、矿物油佐剂疫苗(简称油疫苗)、氢氧化铝胶佐
剂疫苗(简称铝胶疫苗)共5种佐剂的三价灭活疫苗。豚鼠和仔猪安全性试验结果表明:油佐剂疫苗的副反应较
大,其余疫苗的副反应较小。仔猪免疫保护试验结果显示:二免14 d后利用约为1 LD100的SS2 Z2株对仔猪经耳缘
静脉攻毒,ISA201 疫苗、GEL 疫苗对仔猪的保护率均为100%,油疫苗和铝胶疫苗保护率分别为60%、40%,
IMS1313 疫苗组的保护率为0;利用1 LD100 SS1 Z1 株攻毒后,ISA201 疫苗、GEL 疫苗、油疫苗、铝胶疫苗和
IMS1313疫苗对仔猪的保护率分别为100%、80%、80%、60%和0。综上结果表明,ISA201 疫苗免疫仔猪能产生
最高的抗体水平,同时提供了最高的保护效力,且副反应较小,确定ISA201为开发链球菌灭活苗的首选佐剂。
本研究为SS三价灭活疫苗的研发奠定了基础。
Abstract:
The objective of this study was to screen for the immune adjuvant for novel trivalent inactivated vaccine of
Streptococcus suis (S. suis). The trivalent inactivated vaccine was composed of S. suis serotype 1 Z1 strain, serotype 2 Z2 strain
and serotype 7 Z7 strain as antigens, and the immune adjuvants screening here included: ISA201 VG biphasic oil emulsion
adjuvant (referred as ISA201 vaccine), GEL 02 PR water-soluble polymer adjuvant (referred as GEL vaccine), mineral oil adjuvant
(referred as oil vaccine), aluminum hydroxide adjuvant (referred as Alh vaccine) and IMS1313N VG water-soluble nano adjuvant
(referred as IMS1313 vaccine). The safety trial results of guinea pigs and piglets showed that the side effects of mineral oil adjuvant vaccine was most obvious and the rest were little. The immunoprotection trial results of the piglets showed that after 14
days of second immunization, piglets were intravenously challenged with 1 LD100 S. suis 2-Z2 strain, both ISA201 vaccine and GEL
vaccine showed protection rate was 100% meanwhile oil vaccine, Alh vaccine and IMS1313 vaccine showed protection rate was
60%, 40% and 0, respectively. The other group piglets were intravenously challenged with 1 LD100 S. suis 1-Z1 strain, and the
results showed that ISA201 vaccine, GEL vaccine, oil vaccine, Alh vaccine and IMS1313 vaccine provided 100%, 80%, 80%, 60%,
and 0 protective efficacy. Due to high safety, easy of injection, rapidly immune responses, high levels of antibodies, and 100%
protection efficacy for piglets, ISA201 was the best of the 5 adjuvants and could be the optimal adjuvant for the development of an
inactivated vaccine against swine streptococcosis. This study provides the support and reference for the further selection of
adjuvants for inactivated porcine streptococcal vaccine.
