:为了解云南省山羊中阿卡斑病毒(AKAV)的分子遗传特征及自然感染规律,本研究于2019年在云南省
师宗县设立哨兵羊,定期采集血液样品,将核酸检测为AKAV阳性的样品接种BHK-21细胞进行病毒的分离,结果
显示,从哨兵羊分离出3株AKAV(V287/YNSZ/2019、V288/YNSZ/2019和V289/YNSZ/2019),分离株可致BHK-21细
胞出现圆缩、裂解的细胞病变(CPE)。对分离株经电镜观察可见病毒粒子呈球形, 直径在100 nm~
130 nm,表面有纤维状突起。通过PCR扩增其S、M与L基因,并测序与序列分析后分别构建S、M与L基因的进
化树。结果显示分离的3株AKAV的S、M与L基因序列之间高度同源;系统发育分析结果显示,分离株与中国
株亲缘关系最近,与其他亚洲株的亲缘关系次之,与澳大利亚及非洲株的亲缘关系最远;分离株的S及M基因归
属于AKAV基因Ia型,L基因则与中国株共同形成“中国支系”。对定期采集的AKAV感染山羊血液中的病毒核酸
及抗体进行动态监测,结果显示,AKAV核酸在感染动物血液中仅存在一周,而AKAV特异性抗体则可持续9周
以上。本研究首次在中国云南山羊中分离到AKAV,并证实从感染动物血液中分离AKAV的最佳窗口期为病毒感
染后的第1周,对有效开展AKAV的分离工作具有一定的指导意义。
Abstract:
To investigate the molecular genetics and infection characteristics of AKAV pandemic in goats in Yunnan province,
blood samples were collected regularly from the sentinel goats set in Shizong County for the AKAV nucleic acid and antibody
detection, BHK-21 cells were inoculated with AKAV nucleic acid positive samples for virus isolation. Three AKAV strains (V287/YNSZ/2019, V288/YNSZ/2019 and V289/YNSZ/2019) were isolated from the sentinel goats. The isolates induced a cytopathic
effect in BHK-21 cells that involved "rounding up" and exfoliation. The virion was observed through electron microscope. The
virus particles were 100nm to 130nm in diameter, with distributed spikes on the surface. The S, M and L genes of the isolates were
cloned, sequenced and analyzed, then the phylogenetic trees of the S, M and L gene were constructed. Sequence analysis showed
that the sequence of the S, M and L gene of the isolates showed high sequence similarity. Phylogenetic analysis showed that the
isolates shared the closest genetic relationship with the Chinese strains, followed by other Asian strains, and the furthest genetic
relationship with Australian and African strains. The S and M genes of the isolates belonged to the genotype Ia, and the L gene
formed "Chinese branch" with the other Chinese strains. The AKAV nucleic acid and antibody in the blood of the goats infected
with AKAV were monitored weekly. The AKAV nucleic acid was detected in the blood of the infected animals for only one week,
while the specific antibodies lasted for nine weeks at least. In this study, three AKAV strains were isolated from goats in Shizong
County Yunnan province for the first time and the optimal period for virus isolation from the blood of the AKAV infected animals
was the first week after infection. It has certain guiding significance to AKAV isolation.
