为对流行于我国云南省的流行性出血病病毒(EHDV)进行分离与鉴定,以掌握我国流行EHDV的遗传特征与感染特性,本研究将2012年8月采集于哨兵牛的EHDV核酸阳性血液接种BHK-21细胞以分离病毒;通过血清中和试验与分离病毒的Seg-2、Seg-3基因测序分析,确定病毒的血清型与遗传特征;采用C-ELISA和荧光定量RT-PCR(qRT-PCR)方法对EHDV感染哨兵牛血清中的抗体水平与血液中病毒核酸进行动态监测。结果显示,经BHK-21细胞分离出一株EHDV (YNSZ/V003/2012),血清中和试验显示分离病毒属于EHDV-6型。病毒Seg-2、Seg-3基因序列分析显示YNSZ/V003/2012血清型为EHDV-6型,地域型为东方型,与日本EHDV-6亲缘关系最近。哨兵牛感染该病毒后,血清中的特异性抗体迅速上升,感染3周后抗体达最高点并能够在该水平维持较长时间,而血液中病毒核酸含量则呈迅速下降趋势,感染7周后已经检测不到病毒核酸。本研究首次报道了EHDV-6型病毒株在我国云南的分离、序列特征以及在动物中的感染特性,为进一步开展EHDV-6型的流行病学调查和致病性等相关研究奠定了基础。
  
Abstract:
This study aims to isolate epizootic haemorrhagic disease virus(EHDV) that is endemic in Yunnan province of China, and to understand the genetic and infection characteristics of EHDV. EHDV nucleic acid positive blood samples collected from the sentinel cattle in August 2012 was inoculated into BHK-21 cells to isolate the virus. Serotype and genetic characteristics of the isolates were identified by serum neutralization test and sequencing of Seg-2 and seg-3 gene of the virus. Antibody levels and viral nucleic acid in the EHDV infected the sentinel cattle were tested by C-ELISA and qRT-PCR. We isolated an EHDV (YNSZ/V003/2012) from the sentinel cattle. The results of serum neutralization test and sequence analysis of Seg-2 and Seg-3 gene showed that the isolate was serotype 6 EHDV, the regional type was Eastern type, and had the closest genetic relationship with the Serotype 6 strain in Japan. After the sentinel cattle was infected with the virus, the specific antibody increased rapidly, reached the highest point 3 weeks after infection and lasted at this level for a longer time, while the content of nucleic acid in the blood decreased rapidly, and no viral nucleic acid was detected 7 weeks after infection. This is the first report on the isolation, sequence and infection characteristics of EHDV-6 strain in Yunnan province of China. The study provides a basis for further epidemiological investigation and pathogenicity research of EHDV-6 virus.
