摘要:
为了防控A型多杀性巴氏杆菌(Pm)引起的牛纤维素性化脓性肺炎以及B型Pm引起的牛出血性败血症,本研究制备了牛Pm病二价灭活疫苗(A型Pm-TJ株+B型C45-2株),并将疫苗分别以0.3 mL和4 mL接种小鼠和兔后,所有接种动物均健活;分别以单剂量(2 mL)、单剂量重复、超剂量(4 mL)接种肉牛和奶牛后,接种牛均无不良反应,而且接种疫苗对肉牛增重和奶牛产奶量均无影响,表明疫苗安全性良好。以不同的免疫剂量接种牛后,分别采用A型和B型Pm强毒菌液对免疫牛进行攻击,结果显示,疫苗能够对攻毒牛产生良好的免疫保护,并且疫苗的免疫效力呈现剂量依赖性,最小免疫剂量为1 mL (对A、B型Pm攻毒牛的保护率分别达到75 %与100 %)。此外,犊牛接种疫苗10个月后攻毒,对A、B型菌攻毒牛的保护率仍分别能够达到75 %与100 %,表明该疫苗的免疫持续期至少可达10个月。综上所述,该牛Pm二价灭活疫苗能够有效预防牛纤维素性化脓性肺炎和牛出血性败血症,该疫苗的推广应用将对我国牛Pm病的防控发挥关键性作用。
  
Abstract:
To prevent cattle fibrinosuppurative bronchopneumonia caused by Pasteurella multocida capsular type A and hemorrhagic septicemia caused by P.multocida capsular type B, a bovine P.multocida bivalent inactivated vaccine (type A, Pm-TJ strain and type B, C45-2 strain) was prepared in this study and the safety and potency of the vaccine were evaluated in animals. When mice and rabbits were inoculated with 0.3mL and 4mL of the vaccine, respectively, all immunized animals were alive. In addition, when beef calves or cows were inoculated with single dose (2mL), single dose repetition or overdose (4mL), respectively, no adverse reaction was observed in the animals, and the immunizations had no effect on month-weight gain of calves and daily milk production of cows, showing a good safety of the vaccine. In immune potency tests, calves were immunized with different doses of the vaccine followed by challenging with P.multocida capsular type A and type B, respectively. Strong protection was provided by the vaccine for the immunized calves against the challenge in a dose-dependent manner. When the inoculation dose was 2mL per calf, the protection rates of the vaccine against both type A and type B strains were 100%, and the minimum immune dose was 1mL per calf with the protection rates of 75% and 100% against type A and type B strains respectively. Furthermore, calves were still protected against the challenge after ten months of the immunization, and protection rates of the vaccine against both type A and type B strains were still 75% and 100%, respectively, indicating that the vaccine has an immune duration of at least 10 months. In summary, the bovine P.multocida bivalent inactivated vaccine could effectively prevent from fibrinosuppurative bronchopneumonia and hemorrhagic septicemia in cattle, and the promotion and application of this vaccine will play a key role in prevention and control of cattle P.multocida related diseases in our country.
