摘要:
维氏气单胞菌(A.veronii)是严重危害草鱼的一种病原。本研究从电镜观察、Biolog 微生物自动分析仪鉴定、细菌游动能力、胞外产物活性、细胞黏附性、对草鱼致病力和毒力基因等方面,比较草鱼源A.veronii菌株 GZ09007 和 FS12001 的表型特征和致病性差异。结果显示,两株细菌菌体均呈短杆状或弧形、两端钝圆,在透射电镜下可见单鞭毛。Biolog微生物自动分析仪鉴定结果显示菌株均为A.veronii,两株细菌均具有游动能力,胞外产物具有溶血活性、溶蛋白活性和脂肪酶活性,但GZ09007的游动能力、溶血活性、蛋白酶活性均显著强于FS12001 (p<0.05)。GZ09007对EPC、CIK细胞黏附性弱,FS12001 的细胞黏附性强,两株细菌的细胞黏附性差异显著(p<0.05)。菌株GZ09007具有aerA、act、lip、ser、exu、fla毒力基因,但未检测到ast、alt、gcaT、ahyB等毒力基因;菌株FS12001具有aerA、act、alt、ast、lip、ser、ahyB、exu毒力基因,但未检测到gcaT、fla毒力基因。两株细菌均可以造成草鱼发病死亡,GZ09007对草鱼的LD50为5.6×106 cfu,而FS12001的LD50则高于1.5×108 cfu。表明 GZ09007为强毒株,FS12001为弱毒株。推测A.veronii胞外产物的溶血活性和溶蛋白活性在其感染和致死草鱼过程中起到了关键作用。本研究结果为进一步探究A.veronii菌株的致病机理提供依据。
  
Abstract:
Aeromonas veronii is a Gram-negative bacterium and an acute epidemic pathogen of grass carp, causing enteritis and hemorrhage. The two bacteria isolates of GZ09007 and FS12001 isolated from diseased grass carps were identified as A.veronii through Biolog Microbial Identification System. The pathogenicity and phenotype characteristics of the A.veronii isolates were investigated in vitro and in vivo, Virulence phenotype characteristic showed that the two isolates produced hemolysin, protease and lipase. Compared with FS12001, GZ09007 had higher protease activity and hemolytic activity to the erythrocytes of sheep, rabbit, grass carp and silver crucian carp. Compared with FS12001, GZ09007 had quicker swimming motility on 0.3% agar plate, but the adhesion ability to the EPC cells and CIK cells was lower. the PCR amplification of ten virulence genes revealed GZ09007 contained aerA, act, lip, ser, exu and fla, and FS12001 contained aerA, act, alt, ast, lip, ser, ahyB and exu. The in vivo research showed that the whole bacteria GZ09007 had higher pathogenicity for grass carp with the LD50 value of 5.6×106cfu. However, the whole bacteria FS12001 had lower pathogenicity with LD50 value of above 1.5×108cfu. It suggested that, the isolate GZ09007 was highly virulent, but FS12001 was lowly virulent. The result of in vitro and in vivo showed that extracellular protease and hemolysin of A.veronii play a vital role in the pathogenicity. The study provides some basis for further investigation of the bacterial pathogenesis.
