为了解江淮地区猪链球菌(SS)和肠球菌分离株的血清型或基因型分布特征及临床耐药性,本研究对江淮地区临床病例196株分离菌进行SS与肠球菌的鉴定,采用PCR方法进行SS及其1、2、7、9血清型和mrp、ef、sly毒力基因型鉴定;脉冲场凝胶电泳(PFGE)分析SS和肠球菌基因型;K-B法测定其药物敏感性。结果显示,在196株分离菌中,鉴定出112株SS,40株肠球菌。血清型2型SS 44株(39.3 %)为优势血清型,ef -/ mrp-/sly -型SS 36株(32.1 %)为优势毒力基因型,Ps28 18株(16.1 %)和Ps27 14株(12.5 %)为优势PFGE基因型。肠球菌中屎肠球菌21株(52.5 %)为优势菌,Pe12 9株(22.5 %)和Pe7 7株(17.5 %)为肠球菌优势PFGE基因型。SS和肠球菌对20种药物均表现出不同程度的耐药,耐8种以上药物的菌株数分别占85.7 % (96/112)和95 % (38/40)。结果表明,江淮地区SS和肠球菌种型复杂,已经成为引起猪细菌性疾病的重要致病菌。分离菌株PFGE基因型均呈多态性,在传播过程中可能存在遗传变异。SS PFGE基因型与血清型、毒力基因型不呈现相关性。菌株耐药现象严重,多重耐药普遍,且耐药谱越来越广。本研究为江淮地区SS和肠球菌的分子流行病学研究及相关疫病防控提供科学依据。
 

Abstract:
To investigate the serotype or genotype distribution and drug resistance of Streptococcus suis and Enterococcus isolates in Jianghuai area, a total of 196 bacteria were isolated from the clinical pig samples and subjected to distinguishment of S.suis and Enterococcus with full-automatic bacteria analyzer, genotyping of S.suis and Enterococcus by PFGE, identification of S.suis serotypes of 1, 2, 7, 9 and virulence genotypes of mrp, ef, sly by PCR, and determination of drug sensitivity by Kirby-Bauer method. The results showed that there were 112 isolates of S.suis and 40 isolates of Enterococcus. Among S.suis, S.suis type 2 (39.3%) was dominant serotype, ef -/mrp-/sly- type (32.1%) was dominant virulence genotype, and Ps28 (16.1%) and Ps27 (12.5%) were dominant PFGE genotypes. Among Enterococcus, E.faecium (52.5%) was the dominant, and Pe12 (22.5%) and Pe7 (17.5%) were the dominant PFGE genotypes. S.suis and Enterococcus showed different levels of resistance to 20 drugs, and 85.7% and 95% of the isolates were resistant to more than 8 kinds of drugs. It could be concluded that genotypic diversities of Streptococcus and Enterococcus in Jianghuai area were complex, which had become an important pathogenic bacteria causing bacterial disease in swine. The PFGE genotypes of the isolates were polymorphism, and there might be genetic variation in the transmission process, but there were no correlation between PFGE genotypes, serotypes and virulence genotypes of Streptococcus suis. The resistance in the isolates was serious, the multidrug resistance was widespread, and the drug resistance spectrum has been widely dispersed. These data provid a basis of S.suis and Enterococcus for the molecular epidemiology and prevention in Jianghuai area.
