摘要:
为揭示我国猪场中的新发猪繁殖与呼吸综合征病毒(PRRSV)的基因组特征及起源,本研究对辽宁省两个发病猪场检测到的两株1-7-4分支PRRSV (LNWK96和LNWK130)进行遗传演化及测序分析。遗传演化分析显示,两株1-7-4分支PRRSV在我国PRRSV基因分群中属于一个新亚群,即第7亚群。对LNWK96株进行了全基因组序列分析显示,与我国分离的PRRSV株的核苷酸同源性为82.3 %~84.9 %,与美国分离的1-7-4分支IA/2014/NADC34株的同源性最高为96.1 %。重组分析显示LNWK96株是以1-7-4分支的病毒为母本株,ISU30和NADC30病毒株提供重组片段的重组病毒。推测该两株PRRSV极有可能是来自北美的输入性1-7-4分支PRRSV。本研究首次报道1-7-4分支的PRRSV在我国出现,该类病毒株在我国的流行趋势值得关注,其致病性及当前疫苗的保护效果需要进一步研究。
 
Abstract:
To reveal the genomic characteristics and origin of novel emerging porcine reproductive and respiratory syndrome virus (PRRSV), the complete genomic sequences of two novel PRRSV (LNWK96 and LNWK130) detected from two pig farms in Liaoning province were sequenced for the characteristic analysis. Phylogenetic analysis indicated that both PRRSVs belonged to 1-7-4 PRRSVs and formed a new subgroup 7. Complete genome sequences analysis showed that the similarity of LNWK96 were 82.3% to 84.9% compared with epidemic strains isolated from China, and 96.1% identity compared with IA/2014/NADC34 from USA. The recombination analysis showed that LNWK96 derived from recombination of 1-7-4 PRRSVs, NADC30 and ISU30. We speculated that these viruses probably introduced to China from 1-7-4 PRRSVs in America. In the present study, we first reported the emergence of ORF5 RFLP 1-7-4 PRRSVs in China. The epidemic of 1-7-4 PRRSVs should merit the special attention in China, the pathogenicity and the protective effect of commercial vaccines need further study.
