摘要:
为阐明野猪杜氏颚口线虫分离株线粒体细胞色素b (cytb)基因的遗传变异情况,并利用该基因探究杜氏颚口线虫与其它线虫的种群遗传关系。本研究利用PCR扩增从野猪体内分离出的杜氏颚口线虫cytb基因,并对该基因序列进行分析。结果显示,8株杜氏颚口线虫分离株线粒体cytb基因序列长度均为1 067 bp,与Genbank中登录的杜氏颚口线虫分离株同源性均大于98.3 %,与其它线虫的同源性均小于72.0 %;通过该基因建立的系统发育树分析显示,所有的杜氏颚口线虫分离株均与已知杜氏颚口线虫属于同一大分支。表明,线粒体cytb基因序列种内相对保守,种间差异明显,可以作为分子标记应用于杜氏颚口线虫的种间遗传变异研究。本研究为杜氏颚口线虫的分子流行病学和群体遗传学奠定基础。
  
Abstract:
This study aimed to analyze the sequence variations in the mitochondrial cytochrome b (cytb) genes among Gnathostoma doloresi isolates from wild boar in Hunan province, and to reveal its genetic relationships between G.doloresi and other nematodes using cytb gene sequences. The cytb genes of G.doloresi were amplified by PCR and sequenced. The analysis results showed that the length of mitochondrial cytb sequences of 8 isolates were 1,067bp, and the similarities of the corresponding sequences, except Hunan isolates, were more than 98.3% among G.doloresi, and the homology of Hunan isolates and reference of other nematodes strain in GenBank were lower than 72.0%. Through the establishment of phylogenetic tree, the Hunan isolates and G.doloresi available in GenBank were belonged to the same clade. It is concluded that cytb gene sequences could be used as a genetic marker for species genetic variation of G.doloresi. The results of the present study provided a useful basis for further studies of molecular epidemiology of G.doloresi, and diagnosis of the resultant disease.
