摘要:
为了解山东地区鸭坦布苏病毒(DTMUV)的流行及其分子的遗传变异情况,本研究自山东的菏泽、泰安、济宁、枣庄等9个地区搜集临床样品,利用RT-PCR、荧光定量PCR等方法进行检测,并进行分离株E基因及全基因的遗传进化分析;同时,采用阻断ELISA方法检测不同地区未免疫DTMUV病疫苗的不同品种鸭群的血清样品。结果显示,临床样品中DTMUV检出率为13.3 % (36/270)。对分离株E基因的序列分析表明,本研究分离的DTMUV与已发布的禽坦布苏病毒基因的同源性在96.5 %以上。两株分离株的全基因序列分析表明,部分氨基酸位点发生了变异。血清抗体检测结果显示,不同地区及品种的DTMUV感染情况差别较大,肉鸭在德州和滨州未检测到阳性,而在泰安的阳性率为42.2 %,后备种鸭和蛋鸭在各地区的阳性率高低不一;从鸭的品种来分析,蛋鸭是感染最严重的鸭群,阳性率为55.6 %。结果表明,坦布苏病毒对山东水禽养殖仍有一定的危害,目前山东地区流行的DTMUV与之前分离的禽坦布苏病毒同源性较高,未出现明显的分子变异。
 
Abstract:
To investigate the epidemiology and molecular characteristics of duck Tembusu virus (DTMUV) in duck flocks, the clinical samples collected from Heze, Tai’an, Jining, Zaozhuang and other nine regions of Shandong province were detected by RT-PCR and real-time PCRs, while the E genes and whole gene sequence of the isolates were also analyzed. At the same time, duck serum samples from non duck Tembusu (DTMU) vaccine immunized ducks in different regions and different ages was detected by blocking ELISA method. The results showed that 36 positive samples detected from 270 clinical samples and the positive rate was 13.3%. Sequence analysis based on E gene showed that the sequences shared more than 96.5% identity with those of the resent DTMUV isolates. The whole gene sequence analysis of two isolates showed that some amino acid sites were mutated. According to the serum antibody test results of different duck breeds, the infections of DTMUVs were various in different regions and varieties. DTMUV were negative among the meat-type ducks detected in Dezhou and Binzhou, but the positive rate was 42.2% in Tai’an. The positive rate of antibody against DTMUV was uneven among breeding duck and laying duck in every area. From the analysis of breeds of ducks, the most severely infection was found in laying ducks and the positive rate was 55.6%. The results showed that the DTMUV was still threat to the waterfowl breeding in Shandong area. The phylogenetic analysis revealed that no apparent molecular variation was found in DTMUV in Shandong area so far.
