摘要:
为建立产气荚膜梭菌α毒素的双抗体夹心ELISA (DAS-ELISA)检测方法,本研究以抗产气荚膜梭菌α毒素单克隆抗体(MAb)为捕获抗体,抗产气荚膜梭菌α毒素多克隆抗体为检测抗体,建立了检测α毒素的DAS-ELISA。通过优化反应条件,建立标准曲线;并对建立的DAS-ELISA方法进行性能评价及初步应用。采用该方法测定产气荚膜梭菌α毒素的最低检测限为0.353倍的小鼠LD50,可定量范围为0.353~90.368倍小鼠LD50。结果表明该DAS-ELISA方法具有良好的特异性、灵敏性和重复性,可以用于大批量样品检测。初步应用结果显示:患产气荚膜梭菌病的兔群肠内容物中产气荚膜梭菌的α毒素水平显著高于健康兔群(p<0.05)。本研究检测α毒素的DAS-ELISA建立有助于畜禽产气荚膜梭菌病的诊断及研究。
  
Abstract:
In order to establish a method for detection of the Clostridium perfringens alpha toxin, the double antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA) was developed with monoclonal antibody (MAb) and polyclonal antibody against α-toxin as capture antibody and detection antibody, respectively. Under the optimized reaction conditions, the standard curve was established. The performance of DAS-ELISA was evaluated and this method was also preliminarily applied in detection of the clinical samples. The results showed that the assay was specificity, sensitivity and repeatability. The detection range and the detection limit of the DAS-ELISA were 0.353 to 90.368 LD50 and 0.353 times of mouse LD50, respectively, which could be used to high-throughput detect clinical samples. Preliminary application results revealed that the concentration of C.perfringens α-toxin in the intestinal content of diarrhoea rabbits was significantly higher than that of healthy rabbits (p<0.05). In conclusion, the established DAS-ELISA was useful to diagnosis and further study of C.perfringens related disease.
