摘要:
为了解山东省猪流行性腹泻病毒(PEDV)流行情况,本研究采用RT-PCR的方法对2012年1月~2016年12月山东省13个地市采集的258份疑似PEDV感染样品进行检测,并设计引物扩增PEDV ORF3基因片段,通过生物信息学软件对26个具有代表性的完整的PEDV ORF3基因序列进行遗传进化分析。结果显示PEDV平均每年检出率达72.1 %,表明PEDV是当前山东地区仔猪腹泻的主要致病病原。遗传进化分析显示,26株PEDV ORF3基因序列属于G2基因型,与其它参考PEDV病毒株同源性较高(93.8 %~100 %),相对保守。其中3株PEDV流行株位于2a亚型中,这在华北地区属首次报道。PEDV 2a亚型基因序列具有特异位点突变,可作为鉴别2a亚型与其它亚型的分子标记。本研究揭示了山东省PEDV流行株ORF3基因的主要基因型,为该省PEDV的防控提供依据。
 
Abstract:
To investigate the epidemiology of porcine epidemic diarrhea virus (PEDV) infection, a total of 258 porcine intestinal and fecal samples with typical symptoms of PEDV infection were collected and detected by RT-PCR in Shandong province from January, 2012 to December, 2016, and the ORF3 genes of the 26 PEDV positive samples from 13 areas were sequenced and analyzed. Results showed that the average positive rate of PEDV was 72.1% of the 5 years, which indicated that PEDV was the major pathogen for the piglet diarrhea. By phylogenetic analysis of the ORF3 genes, the 26 PEDV were clustered in G2 and revealed high levels of sequence homology with other reference sequences (93.8%-100%). In addition, three of the PEDV were classed into the 2a subgenotype which was first identified in North China. Evidently, there were several mutations in the PEDV ORF3 that could be used for discrimination between 2a and other genotype. This study investigated the molecular diversity of the virus and provided a basis for the prevention and control of PEDV.
