摘要:
为评价分子修饰狂犬病ERA疫苗株(简称rERAG333E)的免疫效果,本实验将其分别以103 FFU/mL、104 FFU/mL、105 FFU/mL、106 FFU/mL的剂量口服免疫小鼠,监测小鼠血清中和抗体持续期;同时在小鼠免疫4周后采用不同现地流行株进行攻毒保护实验。结果表明,rERAG333E以104 FFU/mL以上剂量免疫小鼠,免疫后各实验组小鼠均能够产生较高水平的抗狂犬病病毒(RABV)中和抗体,且能够抵抗经典效检强毒株CVS-24和现地流行强毒株GX/09和MRV的致死性攻击,保护率达到100 %;免疫有效持续期能够达到24周以上。以上结果表明rERAG333E是一种安全有效,具有推广使用潜力的狂犬病口服疫苗候选株。
Abstract:
To evaluate the immune efficiency of the modified ERA strain rabies vaccine (rERAG333E), groups of mice were oral immunized with rERAG333E from the doses of 103 FFU/mL to 106 FFU/mL, and the duration of virus neutralizing antibody(VNA) and protection in mice were detected against the challenging with different genotype rabies virus (RABV) strains at 4 weeks post immunization. The results showed when mice were immunized with more than 104 FFU/mL doses of the rERAG333E, the mice produced high levels of VNA against RABV which persisted more than 24 weeks, and the mice were completely protected from the challenge of classical virulent virus CVS-24 and epidemic virulent virus GX/09 and MRV. These results indicated that the rERAG333E had the potential to serve as a safe and effective oral RABV vaccine strain.
