摘要:
为分析河北地区鸡源致病性大肠杆菌(E.coli)毒力基因的携带情况及其与耐药性的相关性,本研究采用PCR方法和K-B纸片法,测定了2013年~2016年从河北省蛋鸡场分离的104株致病性E.coli的19种毒力基因及其耐药性情况。结果显示,从104株致病性E.coli中检出16种不同的毒力基因,总检出率为84.2 %(16/19),其中毒力基因irp2、fyuA、ompT、iucDa、colv、tsh的检出率均在90 %以上,毒力基因iroN、fimC、papC、hlyF、ECs370、ECs3737的检出率在62.5 %~88.5 %,iucA、astA、vat和iss毒力基因的检出率均低于60 %,Ler、eaeA和sfa毒力基因未检出;E.coli分离菌株对20种抗菌药物呈现出多重耐药性,其中耐11~14种抗生素最多,占总分离菌的50.96 % (53/104),对四环素、阿莫西林、氨苄西林、复方新诺明有较高的耐药率,分别为99.04 %、96.15 %、92.31 %、91.35 %;采用SPSS18.0软件统计学方法分析显示,分离的104株鸡致病性E.coli携带的毒力基因和耐药性两者之间不存在明显的相关性。本研究为河北地区鸡致病性E.coli的防控提供科学依据。
 
Abstract:
In order to investigate the situation of virulence genes and the coreelation of the imulence genes and the drug resistance of the pathogenic Escherichia coli isolates from chicken in Hebei area, we used PCR to detect 19 virulence genes and K-B disc diffusion test to examine drug resistance of the 104 pathogenic E.coli isolated from poultry in Hebei region from 2013 to 2016. The results showed that the total detection rate of virulence genes was 84.2% (16/19). Of which the carrying rate of irp2, fyuA, ompT, iucDa, colv and tsh genes reached up to 90% or more, carrying rates of iroN, fimC, papC, hlyF, ECs370 and ECs3737 genes were 63.5% to 88.5%, carrying rate of iucA, astA, vat and iss genes were lower than 60%, but none of Ler, eaeA and sfa genes were detected. The most frequently observed resistant antibiotics were tetracycline (99.04%), amoxicillin (96.15%), ampicillin (92.31%), chemitrim (91.35%), amikacin (58.33%), chloramphenicol (47.22%) and ciprofloxacin (47.22%). From 2013 to 2016, the isolated chicken pathogenic E.coli had shown multi-drug resistance and increased year by year in Hebei province. These data provide a basis for the prevention and treatment of the pathogenic E. coli caused diseases from chicken in Hebei area.
