摘要:
为了解贵州省不同品种羊TLR2基因序列的多态性特征,本研究以不同品种羊血液DNA为模板,对TLR2基因完整ORF进行PCR扩增、测序及序列分析。结果显示,5种羊TLR2基因扩增长度均为2 355 bp。对贵州黑山羊、黔北麻羊、贵州白山羊、波尔山羊和湖羊5种羊TLR2基因进行种内比对结果显示,贵州黑山羊、黔北麻羊、波尔山羊种内TLR2基因序列保守,贵州白山羊种内A1732T位点存在多态性,湖羊种内在G1133A、T1162G、A1949G等位点存在基因多态性;种间比对结果显示,4种山羊有16个核苷酸位点存在多态性,其中有10个错义突变。4种山羊与湖羊进行比对结果显示,湖羊存在较多的多态性位点,其中有11处位点的差异导致氨基酸的变化。蛋白质结构预测分析显示,不同品种羊TLR2基因呈现的错义突变位点引起了其蛋白质二级结构和三级结构的变化,提示可能存在TLR2参与免疫相关信号通路水平的变化。本研究填补了贵州地方山羊品种TLR2基因多态性空白,为探究TLR多态性与羊疫病易感性关系提供基础研究资料。
  
Abstract:
To investigate the polymorphism of Toll like receptor 2 (TLR2) gene in different breeds of sheep in Guizhou province, the complete ORFs of TLR2 genes were amplified, cloned and sequenced from 5 different breeds of sheep blood samples, and the TLR2 genes were analyzed by the bioinformatics software. The results showed that TLR2 genes of the 5 breed sheep were 2,355 bp. Polymorphism analysis of intra-specific comparison indicated the TLR2 genes of Guizhou black goats,Qianbei Ma goats and Boer goats were genetically conservation. However, the polymorphisms of TLR2 gene were found in Guizhou white goats at A1732T and in Hu sheep at G1133A, T1162G and A1949G. Sixteen single nucleotide polymorphisms(SNPs) were found in inter-specific comparison of 4 breed goats and 10 of them were nonsense mutations. There were many different sites between 4 breeds goats and Hu sheep, and 11 different sites resulted in amino acid changes. Protein structure prediction indicated that those nonsense mutations of TLR2 genes in different breeds of sheep cause the changes of TLR2 secondary and tertiary protein structure and might lead to the change of immune related signal pathways of TLR2. These resultsprovided basic data for further studying the relationship between TLR polymorphism and susceptibility of sheep epidemic disease.
