摘要:
为了解猪胸膜肺炎放线杆菌血清5型(APP-S5)对小鼠的致病性及感染小鼠后的组织分布情况,本研究通过测定APP-S5的生长曲线、筛选APP-S5感染小鼠的最佳接种途径以及观察 不同剂量的APP-S5对小鼠存活的影响之后,选取1.0×107 cfu/只的APP-S5经腹腔注射感染小鼠,以阐明APP-S5感染小鼠后的组织分布规律。结果显示,与肌肉注射和滴鼻相比,腹腔注射接种后小鼠出现的临床症状最早,1.5×107 cfu/只接种小鼠后可于5 h内全部死亡;腹腔感染小鼠后,仅在4 h时肺脏中APP-S5菌体含量最多,而在感染6 h时及8 h时肺脏中菌体含量降低。本研究表明,APP-S5感染小鼠后,具有组织嗜性,且不同接种途径对小鼠的致死性存在差异,本研究结果为深入研究APP-S5的致病性提供了实验依据。
  
Abstract:
To investigate pathogenicity and the tissue distribution in mice infected with serotype type 5 Actinobacillus pleuropneumoniae (APP-S5). After measuring the growth curve of APP-S5, screening the best inoculation route for APP-S5 infected mice, and observing the effect of different dose of APP-S5 on the survival mice. The mice were infected with 1.0×107 cfu/mL dose of APP-S5 by intraperitioneal injection to elucidate the tissue distribution of APP-S5 in infected mice. The results showed that compared with intramuscular injection and intranasal injection, the clinical symptoms firstly appeared in mice by intraperitoneal inoculation, and all the mice died within 5 hours after inoculation with APP-S5 (1.5×107 cfu). When the mice were infected by intraperitoneal, the content of APP-S5 was the highest in the lungs at 4 h after inoculation, but decreased at 6 h and 8 h. Our results demonstrated that APP-S5 had tissue tropism in mice after infected, and the lethal diversities to mice were found in different inoculation routes. Our study provides a experimental basis for further study on the pathogenicity of APP-S5.
