摘要:
为探究无乳链球菌菌毛岛屿3个亚基AP1、AP2、BP融合蛋白的抗原性,本研究根据该3个亚基基因的主要抗原结构域序列设计并合成引物,通过重叠延伸PCR技术获得上述3个亚基的串联核苷酸片段,并插入pET-30a (+)载体,转化至BL21 (DE3)感受态细胞后诱导表达,表达蛋白经亲和层析纯化后进行western blot鉴定和小鼠免疫实验。结果表明,AP1-AP2-BP融合抗原具有良好的反应原性和免疫原性。本研究为进一步研究无乳链球菌基于AP1-BP-AP2融合蛋白的致病机制、检测制剂及亚单位疫苗研制提供了实验依据。
Abstract:
In order to investigate the antigenicity of AP1, AP2 and BP fusion protein in pilus island of Streptococcus agalactiae, primers were designed and synthesized based on the major antigenic domains of the 3 subunits genes. Nucleotide sequences of the 3 subunits were obtained by the SOE PCR technique, and cloned into pET-30a (+) vector to construct recombinant plasmid. The recombinant plasmid was transformed into BL21 (DE3) competent cells to expresse recombiont protein.Purified fusion protein was detected by western blot and immuned mice to preliminarily evaluate the immunogenicity of the fusion protein. The result showed that the AP1-AP2-BP fusion antigen had good reactivity and immunogenicity. This study provides an experimental basis for further study on the pathogenic mechanism of S.agalactiae based on AP1-BP-AP2 fusion protein and development subunit vaccine of Streptococcus agalactiae.
