摘要:
为调查猪源链球菌(S.suis)血清型的流行特点、主要毒力基因的分布及致病力情况,本研究针对来自我国部分地区800份疑似S.suis发病猪的病料样品进行S.suis的分离鉴定、血清型分型、致病力试验及主要毒力基因型检测。研究结果显示,在800份样品中分离到41株S.suis (5.13 %);其中,血清型2型或1/2型S.suis 10株(24.4 %),3型6株(14.6 %),8型3株(7.3 %),9型7株(17.1 %),21型1株(2.4 %),未定型14株(34.1 %);致病力试验结果显示,以1×108 cfu/mL S.suis菌液接种昆明小鼠,41个分离株中具有致死性的菌株19株,其中6株致死性较强,13株致死性较弱;毒力基因检测结果显示gdh、fbps、orf2、gapdh、sly、ef和mrp基因的检出率分别为100 %、82.9 %、75.6 %、41.5 %、39.0 %、19.5 %和12.2 %,以gdh+orf2+fbps+gapdh型(9株)、gdh+orf2+ fbps型(8株)和sly+gdh+orf2+fbps+gapdh型(6株)为主要的毒力基因型。本研究结果揭示了我国S.suis各血清型的流行情况,初步探明S.suis致病力的强弱,为深入开展S.suis防控技术研究及其致病机理奠定了基础。
  
Abstract:
In order to investigate epidemiological characteristics of serotypes, distribution and pathogenicity of key virulence-associated genes of Streptococcus suis (S.suis), this study reported the isolation and identification, serotyping, detection of pathogenicity and key virulence-associated genes of S.suis strains which were isolated from 800 clinical samples of pigs with suspected S.suis infection in partial regions of China. A total of 41 (5.13%) strains isolated from the 800 clinical samples were confirmed to be S.suis, including 10 (24.4%) strains of serotype 2 or serotype 1/2, 6 (14.6%) strains of serotype 3, 3 (7.3%) strains of serotype 8, 7 (17.1%) strains of serotype 9, 1 (2.4%) strains of serotype 21 as well as 14 (34.1%) unidentified S.suis strains. The pathogenicity test was conducted with kunming mice infected with each of the strain at a dose of 1×108 cfu/mL. The result showed that 19 strains were lethal to mice in the 41 S.suis strains, including 6 strains with high lethality and 13 strains with low lethality.
