摘要:
为建立一种用于检测猪圆环病毒3型(PCV3)的PCR方法,本研究针对GenBank中PCV3全基因序列在其保守区设计一对特异性引物,并优化反应条件,建立了PCV3的PCR检测方法。结果显示,该PCR方法仅对PCV3检测为阳性,而对其它常见病原的DNA模板均无扩增。对PCV 3的最低检出量为61.9拷贝/μL。结果表明该方法具有良好的特异性和敏感性。通过该方法对133份临床样品进行检测,结果显示PCV3阳性率为29 % (38/133),表明PCV3在河南地区已有流行。该方法的建立使PCV3的检测更为快速、简便、经济、实用,为PCV3感染的早期诊断提供了有力的技术支持。
  
Abstract:
To establish a PCR assay for detection of PCV3, a pair of special primers were designed according to the highly conserved region of PCV3 complete gene sequences in GenBank. The assay was established by optimizing the amplification conditions. The specific PCR product of 270 bp for PCV3 was amplified by the PCR assay, but no PCR products were aimplified from the other related pathogens. Meanwhile, the detection limit of this method was 61.9 copies/μL for PCV3, which showed that the method had better sensitivity and specificity. The PCV3 positive ratio of 133 clinical samples detected by the PCR assay was 29% (38/133), which indicated the prevalence trend of PCV3 was increased in Henan province. The PCV3 detection method established in this study was characterized to be rapid, economical and easy to handle, which provided effective support for the early diagnosis of PCV3 infection.
