摘要:
 为研究非洲猪瘟病毒(ASFV)主要结构蛋白VP73在哺乳动物细胞中真核表达,本研究根据已发表的ASFV序列,设计合成VP73基因全序列,以巨细胞病毒极早期启动子(CMV-IE)和经水泡性口炎病毒G蛋白(VSV/G)修饰过的pFastBac杆状病毒为载体构建了重组杆状病毒AcNPV-G-VP73。Western blot结果表明,该病毒感染BHK-21哺乳动物细胞后,VP73基因在细胞中获得表达。本研究构建的重组杆状病毒能够介导ASFV VP73蛋白在哺乳动物细胞中表达,为ASF新型疫苗与诊断方法的探索研究奠定了基础。
Abstract:
 To express VP73 protein of African swine fever virus (ASFV) in mammalian cells, the VP73 gene was synthesized and cloned into pFastBac, in which the VSV/G gene is under the control of the polyhedron promoter of pFastBac and the DNA fragment containing the VP73 expression cassette was controled under the CMV promoter (CMV-VP73-BGH poly(A)). In addition, the VP73 protein was expressed in BHK-21 cells infected with the recombinant baculovirus (rAcNPV-G-VP73) generated from baculovirus system and identified by western blot. Our results suggested that VP73 could be used as antigen for further development of vaccine and detection method for ASFV.
