摘 要:为鉴定蓝舌病病毒(BTV)的单克隆抗体(MAb)所识别的VP7蛋白竞争抑制群特异性构象抗原表位,本研究采用噬菌体展示技术对BTV VP7蛋白构象抗原表位氨基酸进行筛选,通过质粒定点突变技术对编码筛选获得的氨基酸区域的基因进行突变,同时利用ELISA法验证该氨基酸区域突变后对BTV VP7蛋白抗原性的影响。结果表明被BTV MAb 4H7所识别的BTV VP7蛋白竞争抑制群特异性构象抗原表位由175FQG177、185YL186和278WH279组成,其中aa175~aa177和aa185~aa186为该抗原表位的关键性氨基酸,本研究初步阐明了BTV VP7蛋白竞争抑制特异性的分子基础,对VP7蛋白功能研究和建立BTV群特异性检测方法具有重要意义。 
Abstract:
Abstract: For identification of conformational epitope in VP7 of bluetongue virus (BTV), the monoclonal antibody (MAb) of 4H7 prepared in our previous study was used to identification of the VP7 protein competitive inhibitory group specific epitope. The conformational epitopes of BTV VP7 were screened with the 4H7 MAb by phage display technology. The results showed that the MAb 4H7 recognized a constituted amino acids of 175FQG177, 185YL186 and 278WH279. In additionally, site-direct mutations based on these amino acids were further confirmed that the aa175-aa177 and aa185-aa186 were critical residues of the epitope. The identification of the competitive inhibitory group specific epitope would facilitate further study on function of VP7 and establishment of BTV group specific detection method.
