摘要:
以纯化的重组PEDV N蛋白为抗原免疫7周龄雌性BALB/c小鼠,利用淋巴细胞杂交瘤技术获得了7株稳定分泌抗重组N蛋白特异性单克隆抗体的杂交瘤细胞株,分别命名为9C1、7H8、3C10、6C4、10G9、2C10和4B3。7株杂交瘤细胞诱导小鼠产生的腹水抗体效价分别为1∶3 200、1∶3 200、1∶6 400、1∶12 800、1∶25 600、1∶64 00和1∶12 800。9C1、7H8、3C10、6C4、10G9和2C10免疫球蛋白类型均为IgM,轻链均为κ链;4B3免疫球蛋白类型为IgG2a,轻链为κ链。Western blot试验结果显示,这7株单克隆抗体均能与天然的PEDV N蛋白发生反应。
Abstract:
BALB/c mice were immunized with 50 μg of purified recombinant N protein of PEDV emulsified in Freund’s adjuvant. Seven hybridoma cell lines 9C1, 7H8, 3C10, 6C4, 10G9, 2C10, and 4B3 that secreted monoclonal antibodies (McAbs) aga- inst recombinant N protein were established. The 9C1, 7H8, 3C10, 6C4, 10G9 and 2C10 were characterised by indirected ELISA as IgM subtype and κ chain, with ascetic titres of 1∶3 200, 1∶3 200, 1∶6 400, 1∶12 800, 1∶25 600, 1∶6 400, respectively. The McAb 4B3 were IgG2a subtype with κ chain and had an ascetic titre of 1∶12 800, Western blot showed that all seven McAbs were able to reacted with native N protein of PEDV.
