利用猪流感病毒(Swine influenza virus,SIV)与伪狂犬病病毒(Pseudorabies virus,PRV)血清学阴性的4周龄断奶仔猪,对此前构建的表达SIV A/Swine/Inner Mogolian/547/2001(H3N2)血凝素(HA)基因的重组伪狂犬病病毒(rPRV-HA)进行了免疫效力评价。按每头105.0 PFU rPRV-HA肌肉注射断奶仔猪(n=12),同时设Bartha-K61免疫对照组(n=5)和非免疫攻毒对照组(n=5)。免疫后35 d用2×105.0 TCID50的SIV A/ Swine/Heilongjiang/74/2000(H3N2)进行强毒攻击。rPRV-HA免疫组免疫后7 d均可检测到高滴度针对SIV的血凝抑制(HI)抗体,而所有对照组小猪仅在攻毒后7 d才开始产生针对SIV的HI抗体;rPRV-HA免疫组在病理学保护方面明显优于对照组,支气管和肺泡冲洗液病毒分离结果显示,攻毒后7 d rPRV-HA免疫组没有检测到病毒,而两个对照组可检测到病毒。以上试验结果表明,rPRV-HA免疫猪对同亚型SIV的攻击具有很好的保护作用。
Abstract:
A recombinant pseudorabies virus (rPRV-HA) expressing the hemagglutinin (HA) gene of swine influenza virus (SIV) A/Swine/Inner Mogolian/547/2001 (H3N2) was immunologically evaluated in piglets. Twelve 4-weeks old healthy piglets were inoculated intramuscularly (i.m.) with 105.0 PFU of rPRV-HA. Two control groups (five piglets each) were either mock-inoculated or inoculated with PRV Bartha-K61. Piglets were challenged intranasally with 2×105.0 TCID50 of A/Swine /Heilongjiang/74/2000 (H3N2) (SwHLJ74) at 35 days post-inoculation. Serological detection showed that piglets inoculated with rPRV-HA developed a high level of virus-specific hemagglutination-inhibition (HI) antibody at seven days post vaccination. In comparison, pigs in the two control groups only developed virus-specific HI antibody at seven days post challenge. Anti-PRV neutralizing antibodies were detected at seven days post vaccination in the sera of all piglets inoculated with rPRV-HA and Bartha-K61. Piglets immunized with rPRV-HA were shown protected from histopathological effects. Compared to the control group, SIV in bronchus and lung lavage fluids of rPRV-HA immunized pigs could be detected for a much shorter time after challenge. Thus, the rPRV-HA can be a potential vaccine candidate for preventing the same subtype of SIV infection.
