摘要:
为研究猪流行性腹泻病毒(PEDV)非结构蛋白1 (NSP1)在Vero E6细胞中的分布和亚细胞定位情况,本研究采用RT-PCR方法扩增NSP1基因,并将其克隆至真核表达载体pAcGFP-C1中构建重组质粒pAcGFP- NSP1。将pAcGFP-NSP1转染至Vero E6细胞中进行瞬时表达,并收集转染后48 h的细胞进行western blot检测。通过激光共聚焦显微镜观察NSP1在细胞中的定位情况。结果表明,NSP1在Vero E6细胞中表达,其主要定位于细胞质中,与细胞中的线粒体、内质网、高尔基体均呈现高度共定位。本研究在Vero E6细胞中表达了PEDV NSP1蛋白,并首次对其亚细胞定位进行了解析,为进一步了解病毒的复制及其致病机制的研究提供了依据。
  
Abstract:
To study the localization of porcine epidemic diarrhea virus (PEDV) non-structural protein 1 (NSP1) in the cells, the PEDV NSP1 gene was amplified by RT-PCR and cloned into eukaryotic expression vector pAc-GFP-C1 to construct the recombinant plasmid pAc-GFP-NSP1. Vero E6 cells were transfected with the pAc-GFP-NSP1 and the subcellular localization of the expressed NSP1 was analyzed by confocal microscopy. The results showed that the NSP1 localized in mitochondrion, endoplasmic reticulum and Golgi apparatus of the transfected cells, suggesting NSP1 had the ability of localization cytoplasm, which provided the basis for further studies of PEDV replication and pathogenesis mechanisms.
