摘要:
为探讨猪圆环病毒2型(PCV2)作用下淋巴细胞的免疫活性,本研究通过体外培养仔猪脾脏淋巴细胞及巨噬细胞(Mφ),分别以PCV2和PCV2+Mφ与淋巴细胞相互作用,同时设Mφ和空白对照组。培养0h、6h、12h、24h后收取各组淋巴细胞,放射免疫法检测胞内cAMP和cGMP浓度。另外,选取15头PCV2阴性仔猪,每头滴鼻PCV2病毒悬液,分别于14d、21d、35d迫杀,每次5头,并设立阴性对照组4头。放射免疫法检测脾脏cAMP和cGMP浓度,并计算cAMP/cGMP比值。体外检测结果显示:PCV2对体外培养淋巴细胞内cAMP和cGMP浓度影响不大;而在Mφ存在时PCV2在6h时能显著降低cAMP和cGMP浓度和cAMP/cGMP比例(p<0.05);12h时cGMP浓度下降(p<0.05),cAMP和cGMP比例上升(p<0.05);24h时cGMP浓度上升(p<0.05),但cAMP/cGMP比例降低(p<0.05)。体内检测结果表明,PCV2能显著降低仔猪脾脏细胞内cAMP浓度(p<0.05)及cAMP/cGMP比值(p<0.05)。该研究结果表明,在巨噬细胞存在时PCV2能够降低淋巴细胞内cAMP/cGMP的比值,从而影响淋巴细胞的功能。
Abstract:
To investigate the immunocompetence of lymphocytes induced by porcine circuvirus type 2 (PCV2), lymphocytes and macrophages (Mφ) were isolated from porcine spleen and the lymphocytes were divided into 4 groups including lymphocyte control group, PCV2 inoculated lymphocyte group, co-culture with Mφ with or without PCV2 inoculation (PCV2+Mφ or Mφ) group. The cAMP/cGMP concentrations were measured by radioimmunoassay. The results indicated that the cAMP/cGMP concentration and cAMP/cGMP ratio was decreased in lymphocytes/PCV2+Mφ group (p<0.05) at 6 and 12 hours, and the cAMP/cGMP ratio at 12 hours, cGMP concentration and cAMP/cGMP ratio at 24h were also increased, comparing with the control and PCV2 inoculated lymphocyte groups (p<0.05). Otherwise, fifteen piglets were inoculated with PCV2, with 4 piglets as negative control, and the piglets were sacrificed at 14, 21 and 35 days post inoculation, respectively. The cAMP/cGMP in spleens was evaluated by radioimmunity. The results showed that the concentrations of cAMP in speen cells of PCV2 inoculated piglets were lower than control group (p<0.05), and cAMP/cGMP ratio was also decreased. In conclusion, PCV2 could change the concentration and the ratio of cAMP/cGMP in lymphocytes via macrophages
