对从广东地区流行鸭肝炎病例中分离到的1株病毒(命名为“GD株”)进行了鸭胚(DE)传代培养。DE2~DE6代鸭胚毒的ELD50在10-5.17/0.2 mL ~10-6.38/0.2 mL之间。DE6代毒回归雏鸭后可以产生与1型鸭肝炎病毒(DHV)相同的临床症状和解剖病变。鸭肝组织毒对4日龄、8日龄和16日龄雏鸭的LD50分别为10-6.5/0.5 mL、10-5.38/0.5 mL和10-4.83/0.5 mL。病毒对氯仿、乙醚和胰酶不敏感,能够耐受pH3.0酸处理,62 ℃ 30 min不能被完全灭活。病毒核酸类型鉴定为RNA病毒。生物学和理化学特性鉴定结果表明,GD株为强毒DHV。使用标准1型DHV阳性血清进行的交叉中和试验和交叉被动保护试验表明,GD株病毒在抗原性上与1型DHV无关。对DHV抗原相关的VP1基因序列分析和同源性比较表明,GD株与DHV-1的VP1核苷酸序列同源性为69.82 %,氨基酸序列同源性为71.77 %。GD株属于一种在我国流行的新型DHV 强毒(暂命名为“CN-DHV”)。
Abstract:
A virus strain (designated as GD strain) was isolated from dead ducklings with duck hepatitis in Guangdong province. The virus could cause death over 80 % in 10-11 day-old duck embryos when inocubatied with 100 ELD50 per 0.2 mL. Intramuscular injection of duck embryos viruses to ducklings caused same clinical symptoms and pathological changes as the type 1 duck hepatitis virus (DHV-1). The virus was not sensitive to chloroform, diethyl ether and trypsin and possessed acid-resistant and heat-resistant features. The GD strain belongs to a virulent DHV based on biological and physicochemical characteristics, but was not related to DHV-1 as shown by in vitro cross-neutralization assay and in vivo cross-protection test with DHV-1 antiserum. Sequence comparison showed that the nt and aa sequences of VP1 of GD strain and DHV-1 shared identities of 69.82 % and 71.77 %, respectively. Therefore, GD strain represents a new serotype of DHV occurred in China (designated CN-DHV).
